Loratadine is an efficacious, non-drowsy antihistaminic molecule prescribed for allergic skin. The conventional oral dosage form associated with various side effects, limited and variable absorption. present study attempts to improve the effectiveness of drug by formulating a niosome loaded hydrogel loratadine treating Ether injection method was used prepare niosomes using Box Behnken design. Particle size analysis, surface charge, morphology (atomic force microscopy), compatibility (Fourier transform infrared spectroscopy), thermal (differential scanning calorimetry), characteristics (powder X-ray diffraction) were done evaluate drug-loaded niosomes. optimized formulation incorporated into two types gelling systems. nanogels evaluated rheological mechanical properties, in vitro, ex vivo permeation skin irritation studies. design found be productive nano-sized (109.99 nm) stable -37.5 mV loratadine. atomic microscopy revealed formation unilamellar niosomes, no incompatibility detected between excipients through Fourier spectroscopy study. differential calorimetry powder diffraction reported confinement drugs niosomal core. had good properties. maximum penetration from gel base Xanthan gum, nanogel. evaluation have satisfactory can promising alternative approach administration treatment allergy.
